     本产品是采用胶体金免疫层析技术快速检测小鼠抗体亚型的测试卡，5分钟快速显色，可用于检测小鼠抗体的重链和轻链亚型，检测的样本包括杂交瘤上清液和纯化的小鼠抗体，不建议直接用腹水作为检测样本，但本产品仍然可用作鉴定腹水中主要抗体亚型的分布。

使用方法

    1. 打开包装取出检测卡，平放在实验台上；

    2. 用微量移液器吸取80~100μl样本，滴加到加样孔中；

    3. 静置5~10min读取结果，15min后判读结果无效。 

结果分析

    1.无效结果：未出现“C”线；

    2.阴性结果：只出现“C”线；

    3.阳性结果：

         a）单克隆抗体：除“C”线之外，只出现一条重链线和一条轻链线。

         b）多克隆抗体：除了“C”线之外，出现多条重链线和多条轻链线；根据红线的深浅可以判定主要亚型。

性能

    最低检出浓度为100ng/ml，各亚型之间无交叉反应。

储存

   常温，保质期18个月。

 注意：

    1. 如果组织培养基的颜色导致高背景，可追加40~60μl PBS以清除背景；

    2. 试剂卡不建议冻存；

    3. 本检测卡仅限于科研使用。

Background

The Rapid Mouse Immunoglobulin Isotyping Kit is a five-minute lateral flow assay with ELISA sensitivity for monoclonal antibody class and subclass determination. Acceptable sample types include hybridoma culture supernatant and purified monoclonal antibody. Ascites fluid is not recommended since it usually contains multiple immunoglobulins. However, this kit can still be used to determine the relative distribution of the main isotypes in a sample of ascites fluid.

Add 100 µl of mouse antibody solution at appropriate concentration into the sample well. Within 5 minutes, a test line (T) will appear showing the isotype of the antibody. A control line (C) will appear at the far end, showing a successful run. This assay is faster and more convenient than the traditional ELISA method.

The isotyping kit has three different sizes with 5, 10 or 20 pouches containing two cassettes per pouch: one cassette is for determining IgG1, IgG2a, IgG2b and IgG2c, and the other cassette is for determining IgG3, IgM, κ and λ. Both strips contain a control line “C”.

While BALB/c mice express IgG2a, some other inbred mouse strains such as C57BL/6, C57BL/10 and NOD do not have the gene for IgG2a and instead express the IgG2c isotype.
